Tetracarpidium conophorum seed extract reduces intestinal absorption, and increases cellular trapping of glucose

Abstract Background Tetracarpidium conophorum is one of the numerous folklore medicinal plants for managing diabetes but mode action and bioactive compounds responsible antihyperglycemic property are missing in literatures. This study aimed at investigating possible modes its using both in-vitro ex-vivo methods. Powdered seed (TECOSE) was extracted with methanol standard extraction procedure. Gas chromatography- Mass spectrometry (GCMS) analysis extract, effects on tissue glucose uptake, α-amylase, α-glucosidase glucokinase enzymes were assessed laboratory procedures. Results Seven heterocyclic identified by GCMS which structurally related to sulphonylurea. TECOSE strongly inhibited (IC 50 = 1.90 mg/ml) partially α-amylase 7.20 activities. Also, activity uptakes significantly ( p < 0.05) increased TECOSE. Conclusions The results obtained deduced that could be due modulation postprandial hyperglycaemia through inhibition intestinal α-glucosidase, increasing activity, improving peripheral uptake mimicking sulfonylurea action.
